I received a substantial check three years ago from a guy for whom I worked. I recently tried to deposit it, but it came back as non-sufficient funds. Do I still have any rights for collecting the funds?
That will depend on whether he still owes you the money. If you had a contract with him, it's likely that it's still enforceable if only three years has passed, but don't sit on the matter any longer. Contact him and tell him you finally tried to cash the check, but it bounced, since he probably thought you had forgotten about it.
Then ask him to give you another check (swap checks with him in person if you can) that you can cash without being concerned that it's so old.
